How do I include supplier shipping?

If you absorb it, put it in shipping absorbed. If the customer pays it separately, leave it out — the calculator prices only what you sell for.

Do I need wastage for POD?

Usually not, since the supplier handles printing. Set wastage above zero only if you routinely eat costs for misprints the supplier does not refund.

Is wholesale realistic for POD?

Rarely. POD base costs are already high relative to bulk printing, so a 40 to 50% wholesale discount often leaves nothing. The calculator confirms this quickly.
